I made some graphics for an RPG Maker project. Do you think I could adapt them to be used in IceBlink? Genre-wise it is a better match to what type of game I want to create.

Being that the graphics are 2.5D, they should work fine on a square grid map. Keep in mind that IB shows you to use either tile based maps and/or large single image maps (which the toolset will split up the large single image into tiles and place in a folder for you).

Can images be larger than 1 tile? For instance, a tree that is 2 tiles high? Or a monster that is 2x2 tiles wide?

Tiles are only one square, but some builders have split tiles into two...Pongo for The Elderin Stone created trees that cover two squares by having two tiles (one for trunk and one for top of tree). Props can be larger than one square. Creatures can be 1x1, 2x1, 1x2, or 2x2 squares.

I would like to walk behind the tree however. I.e. the top part of the tree should not block movement.

It's no big deal. IB is a neat project, but it might just not be for me.
